Generally speaking, in the tissue culture production process of plant factories, the pollution rate should be controlled within 5%. However, in actual production, it is very common that the pollution rate exceeds 5%, and some factories even exceed 10%. The higher the pollution rate, the higher the rejection rate of the tissue culture seedlings produced, which leads to a decrease in the quantity and quality of finished tissue culture seedlings, and the production cost of the factory also rises. So, where does the pollution come from?

ONE  Plant tissue culture and clean air

Before that, we first need to understand what plant tissue culture is.

Plant tissue culture refers to the cutting of organs, tissues or cells from plants. Aseptic culture is carried out on the artificially configured culture medium, and at the same time, by controlling the composition of the culture medium and other environmental factors such as temperature and light, the culture is directed to grow and differentiate to the process of developing and forming a complete plant. If the pollution is not well controlled during the period, it is easy to cause microbial infections such as bacteria/fungi in the culture, unable to grow and develop normally, and even die.

According to research findings, most of the pollution sources (fungi/bacteria) appearing in plant tissue culture are derived from dust particles in the air, except for the bacteria carried by the explants and the incomplete sterilization of the medium/inoculation tools.

① Inoculation operation

During the inoculation operation, if the cleanliness of the air in the tissue culture room is not up to the standard, or the air filtration of the ultra-clean workbench is not thorough, these dust particles and microorganisms floating in the air will land when the operator comes into contact with the plants. It multiplies on plant tissues and culture media, causing tissue culture seedling pollution.

② Nurturing process

During the cultivation process of tissue culture seedlings, if the cleanliness of the cultivation room is poor, the airflow is fast, causing dust particles to move around, or the personnel entering the cultivation room have not been thoroughly sprayed and brought more mites and dust on their bodies, which will also cause the Cultivation pollution.
